Abstract

ABSTRACT

Aim: Glioblastoma multiforme (GBM) is one of the most aggressive brain tumours known for its high-grade malignancy and development of resistance to treatment. In this study, the effect of the combination of Allium polyanthum extract, known for its anticancer potential, and the chemotherapeutic agent dosotaxel on histone deacetylases (HDACs) in GBM cells was investigated.

Method: HDACs, which act as epigenetic regulators, play an important role in tumour cell proliferation and apoptosis processes. In the study, different concentrations of Allium polyanthum extract, dosotaxel and their combinations were applied to glioblastoma cell line. Cell viability was evaluated by cytotoxicity tests and HDAC gene expressions were analysed by real-time PCR (qRT-PCR).

Results: It was observed that the combined treatment resulted in a significant decrease in HDAC1 and HDAC3 expressions, indicating that tumour suppressor mechanisms may be activated at the epigenetic level.

Conclusion: The findings suggest that co-administration of Allium polyanthum with dosotaxel may suppress the growth of glioblastoma cells through HDACs and may be a potential supportive treatment strategy.
